I don't think so, but there's a bus that can take you to Aldershot, where you can transfer to the train to take you to Union.
If you take the 10, 51, I think 1, and probably a few other HSR bus routes, you can easily and quickly get to Hamilton GO station. From there you can get the 16 GO bus, which is the express to Union.

Andrew A is wrong, there is. No need to go to Aldershot. It's the 47G GO bus. You can find their schedule here.

In addition, any bus heading East on Main St. or King St. around McMaster goes to the Hamilton GO station.

47G goes from Hamilton GO to Yorkdale GO on the weekends. There isn't a "direct" bus -- I don't know if that's what you mean by express -- to Union GO from McMaster.

QEW express (16, departs hunter street go station) is the only one I know of... I'm not sure the 47G is an express..? Theres also Lakeshore West (train) but that's not at all quick, or comes at predicable intervals.

I don't think they're direct/express.
If you look at the legend, it says that trips marked with an 'A' require a transfer at Aldershot and those marked with a 'B' require a transfer at Burlington.

There's the 16 from Union, that doesn't take too long. If you get off at Main and Paisley (just after you get off the highway) you can easily walk to campus (would take about 15ish minutes.)

That looks like the 16's schedule. According to the GO mobile app, there is no bus that departs McMaster university and goes directly to Union. Buses at McMaster go to... Aldershot, Bramlea, Square One, York...
You can either go to hunter street GO (or dundurn at king) or go to one of those above places and transfer.

The ONLY express bus that runs between Toronto and Hamilton is the QEW. The 47 has many stops and takes about half an hour longer, and does not go to Union Station.
The fastest way to access this bus, is to take the 5A/51/1A from McMaster to either Main at Dundurn, and walk up to King at Dundurn where the bus stop is. Or you can take those buses to Main at Hughson, and walk down to Hamilton GO Station and get the bus from there.
The 51 goes straight into the Hamilton GO Station.
Coming from Toronto, the bus doesn't go straight to McMaster, but the first stop is about ten blocks away from the university, and you can catch a bus on Main street that will take about two minutes.
