QEW Express: http://www.gotransit.com/publicroot/...&station=&new=
You will have to get off at Columbia College (Main and Longwood) and walk to McMaster, it's approximately 10-15 minutes)
Train and Bus: http://www.gotransit.com/publicroot/...&station=&new=
You end up on campus with this route.

There's the Lakeshore West train from Union station to either Aldershot or Burlington Station, followed by a bus to McMaster. However, I'm not sure if this is still running in the summer :/.
Your best bet is the QEW bus from Union Station to the Hamilton GO centre .

Other direction, the OP wants to end up at McMaster XD The Train/Bus only runs on weekdays during the summer.

No, it runs on weekends too...the bus from Aldershot goes to Columbia College and Main & Longwood (these are actually two separate stops, although it shows up as one stop on the schedule because they're so close) on its way to Hamilton Go.

So... it still doesn't run from the McMaster campus on weekends in the summer. You would end up in the same place if you were to take the QEW express, the only difference is with the QEW express you don't have to transfer.
